Investigation of PK08 (synthetic toltrazuril derivative) toxicity and recommended dosage for olive flounder (paralichthys olivaceus)

Olive flounder, which occupies for more than 50% of Korea's farmed fish production, has recently been damaged by K. septempunctata infection, a myxospore of the genus cudoa. Due to this food safety problem, there are concerns about a decrease in exports and domestic consumption of olive flounder in Korea. Toltrazuril was developed in the 1980s for the purpose of treating and preventing parasitic diseases in livestock. So far, it has been approved as veterinary drug and is widely used in the livestock (Mehlhorn et al., 1988). In the acute toxicity test using toltrazuril on olive flounder, the lethal dose 50% (LD50) of olive flounder was about 45 times stronger than that of rat. Synthetic toltrazuril derivative, PK08 has been developed that can reduce toxicity through structural changes of toltrazuril. Based on the results of range-finding tests, it was found that PK08's no observed adverse effect level (NOAEL) for olive flounder was 120 mg/kg, the lowest observed Adverse Effect Level (LOAEL) was 150 mg/kg, and the definite toxic dose was 200 mg/kg. As a result of the immunotoxicity test, the 200 mg/kg experimental group was close to LD50, so predictable toxicity findings were found, and these results can be used as a reference to determine the effective concentration that can be used for the treatment of olive flounder infected with K. septempunctata.
