Dictyota coriacea and Agarum cribrosum ethanol extracts inhibit melanin synthesis and collagenase activity in artificial skin

With the aim of reducing or replacing animal testing, many in vitro methods have recently been provided as alternatives to using animals. A variety of human skin equivalents have been developed as alternatives to animal testing, and some of them are commercially available. Human skin equivalents can be divided broadly in to two categories. One is an epidermal model containing only normal human keratinocytes cultured to form a multilayered epidermis, such as EpiDerm (MatTek, MA, USA) and EpiSkin (SkinEthic, France). Melanin is produced in melanocytes located in the basal layer of the epidermis of skin, and is a major determinant of skin color. Melanin accumulation in keratinocytes can make skin darker. Although melanin pigmentation in the skin is a protective mechanism against UV radiation, excessive accumulation of melanin induces hyperpigmentation and even pigmentation disorders such as melasma, freckles, and senile lentigines. The synthesized melanin is transferred to keratinocytes by melanocyte dendrites. Melanin accumulation in keratinocytes can make skin darker. Therefore this study aimed at identifying novel cosmetic ingredients as the development of cosmetic products from novel bioresources mushrooming. Edible seaweed has been consumed and used in traditional medicine for centuries by the east ancient people. In the present study, the anti-melanogenic and anti-wrinkle properties of seaweed extracts from Dictyota coriacea and Agarum cribrosum on the artificial skin were aimed to outline. The result of Fontana-Masson stained artificial skin (Neoderm-Me) revealed that the treatment with F1-T-F17 (Dictyota coriacea) and EA-F4-g (Agarum cribrosum) fractions of seaweed extracts significantly decrease the melanin contents by 60%. And IHC (immunohistochemitric) staining of artificial skin (Neoderm-ED) treated with F1-T-F17 and EA-F4-g fractions of seaweed extracts significantly impacted on wrinkle formation. Collectively this study confirms the applicability of Dictyota coriacea and Agarum cribrosum extracts in novel cosmeceutical formulations.
