Soybean is a main crop in terms of economic and nutritional aspects, containing rich amounts of protein, starch, essential minerals as well as secondary metabolites. Soybean core collection used in this study consisted of more than 400 soybean resources and well-reflected more than 99% genetic diversity of 26,000 soybean accessions. Using this core-collection, new metabolites and bioactive compounds can be found based on metabolite-bioactivity association study without traditional bioactivity guided fraction, which requires long and repeating fraction procedures. Based on this platform, biological activity of DPPH radical-scavenging assay was performed to obtain IC50 value with six different concentrations. Meanwhile, hepatocyte protection assay was also carried out to find most effective soybean variety. Metabolite profiling data were obtained based on liquid chromatography high- resolution mass spectrometry (LC-HRMS) and more than 100 metabolite features were used for the association study. After obtaining the profiling of metabolite and bioactivity values, data analysis based on correlation analysis was conducted using a Pearson correlation by R program and showed strong association in between 16 metabolites and bioactivities. The results showed that Cyanidin-3-O-glucoside, Peonidin 3-glucoside, and procyanidin B2 had high correlation with biological activity with more than 0.7 correlation constant. Soybeans rich in Cyanidin-3-O-glucoside, Peonidin 3-glucoside, procyanidin B2 and epicatechin showed both high antioxidant activity and high hepatocyte protection activity which were already indicated by the association study. This study demonstrated that functional metabolomic platform by an association study between metabolite and bioactivity based on core-collection can be an efficient way to find bioactive compounds applicable for various bioactivity including as anti-cancer, anti-diabetes, and many others.
