If the function of salivary glands that secrete saliva is impaired, saliva secretion will decrease and symptoms of xerostomia will appear. Although symptoms of xerostomia are often felt by diabetes mellitus patients, the relationship between diabetes and xerostomia is currently unclear. The objective of this study was to investigate whether apoptosis in salivary glands of diabetic mice might be altered based on immunohistochemistry staining. Immunohistochemistry and Western blot analyses were performed using Fas, Bax, and cleaved caspase-3 antibodies. TUNEL assay was also performed. As a result, expression levels of apoptotic signal molecules of Fas, Bax, and cleaved caspas-3 were increased in DM group. The number of TUNEL positive cells was also increased in DM group. Therefore, xerostomia, a complication of DM, suggest to be caused by apoptosis.

침을 분비하는 침샘의 기능이 손상되면 침 분비가 감소하여 구강건조증의 증상이 나타난다. 당뇨병 환자에게서 종종 구강건조증 증상이 나타나지만, 당뇨병과 구강건조증 사이의 관계는 명확하지 않다. 본 연구에서는 db/db 마우스의 침샘에서 세포 사멸이 나타나는지 확인하였다. Fas, Bax 및 cleaved caspase-3 항체를 사용하여 면역조직화학염색과 Western blot 분석을 수행하였으며, TUNEL 염색도 수행하였다. 그 결과 Fas, Bax 및 cleaved caspas-3 의 세포 사멸 신호 분자의 발현 수준은 DM 군에서 증가 하였다. 또한 TUNEL 양성 세포의 수 또한 DM 군에서 증가하였다. 따라서 당뇨병의 합병증인 구강건조증은 침샘 분비세포의 세포사멸에 의해 유발되는 것으로 사료된다.
