초록/요약

The purpose of this study was to investigate the bacterial diversity in water discharged from dental unit waterlines (DUWLs) using pyrosequencing and to verify the biofilm formation, binding, extracellular polysaccharide production, and coaggregation abilities of the bacterial isolates from these DUWLs. In addition, the present study examined the susceptibility of the DUWL isolates to various disinfectants and confirmed the effects of the disinfectants at susceptible concentrations on multispecies DUWL biofilms formed in the laboratory model. The water samples were collected from 15 dental units located in three different dental clinics in Gangneung city, and four of the 15 samples were selected for pyrosequencing analysis. Isolates from 12 of the 15 water samples were tested for their biofilm formation, binding, extracellular polysaccharide production and coaggregation abilities. The minimum inhibitory concentration (MIC), minimum bactericidal concentration (MBC), minimum biofilm inhibitory concentration (MBIC), and minimum biofilm eradication concentration (MBEC) of the disinfectants against these isolates were recorded. The multispecies DUWL biofilm formed using the CDC biofilm reactor was treated at MBIC and the current recommended concentration of the disinfectants for determining the disinfection efficiency. A total of 233 bacteria were obtained from the four pyrosequenced water samples. The most frequently found genera were Sphingomonas (25%) and Halomonas (20%). Based on biofilm accumulation, 12 of the 56 isolates from the 15 water samples were classified under strong, moderate, or weak biofilm-producing groups. The binding ability of the isolates belonging to the strong biofilm-producing group was higher than that of isolates from the other groups. Susceptibility of these 12 isolates was lowest to ethanol and highest to cetylpyridinium chloride. Similar to the effect of disinfectants at the currently used concentration for DUWL disinfection, the use of cetylpyridinium chloride at MBIC or at 1/2 of MBIC in combination with chlorhexidine against multispecies DUWL biofilms exhibited high disinfection effect. The results of this study will help to understand the mechanism of biofilm formation in DUWL and provide basic data required for developing effective disinfection methods.
