Carotenoid profiling of genetically diverse 355 peper varieties using mass sepctrometry and the association study with antioxidant activity Jin Ho Jo Department of Marine Molecular Bioscience Grauate School GangneungWonju National University Ⅰ. Abstract Phytochemicals are not necessary for life but they help to promote optimal health by lowering risk for chronic diseases, such as cancer and heart disease. They are found only in plant. Phytochemicals are listed as secondary metabolites that are naturally found in plants with roles involved in the restoration of damaged cells, but also in determination of colour, aroma and taste of the plants. These types of products are classified based on the starting point of their biosynthesis: phenolic compounds, carotenoids, products with nitrogen and alkaloids. Carotenoids are a natural dye belonging to phytochemical and synthesized by various resources including, plants, vegetables, fruits, and some microorganisms. Carotenoids have been widely used in the food industry because of its beneficial role in the prevention of diabetes, cancer, cardiovascular, chronic disease. Current HPLC-DAD for carotenoid analysis have been commonly used, but limited analytical application due to their poor sensitivity and selectivity. Liquid chromatography-APCI tandem mass spectrometry has been reported to be efficient for carotenoid analysis through the efficient ionization of these compounds with good selectivity. We have developed and optimized HPLC-APCI-tandem MS/MS for the analysis of targeted 12 carotenoid compounds, including, beta-carotene, neoxanthin, capsorubin, violaxanthin, capsanthin, zeaxanthin, lutein, beta-cryptoxanthin, lycopene, antheraxanthin, phytoene, neurosporene. After successful optimization for the targeted carotenoid compounds. We applied the method to obtain metabolite profiling of 355 pepper accessions which are genetically diverse. ABTS antioxidant activity was also performed for all 355 pepper accessions to figure out how strongly genetic dives of pepper accessions are associated with metabolic and antioxidant variations. To investigate the correlation between the normalized value of carotenoid content and the degree of antioxidant activity in normal red pepper, SIMCA software showed that Neoxanthin and beta-carotene among the 12 carotenoids had higher contribution than antioxidant carotenoids Can be.
