Study on Purification of Paralytic Shellfish Toxin from Toxic Blue Mussel

The official analytic method to determine the paralytic shellfish toxin (PST) amount in Korea is still mouse bioassay (MBA) that has low sensitivity and experiment error. So, most of counties are using instrumental analysis (HPLC, LC/MSMS, etc.) instead of MBA method for analysis of PST. Therefore, it is necessary for the development and analysis of the equipment sanitary safety of shellfish industry. The standard toxin of PST are necessarily needed for instrumental analysis of PST. The purpose of this study was to develop the purification method of PST from toxc blue mussel (Mytilus edulis) was collected at Duck-dong, Masn and Sibang, Keoje, Gyunsangnam-do, and Yongho-dong, Busan, Korea, in May, 2015. The main toxin components in toxic blue mussel were GTX1, 2, 3 and 4 but amount of STX was little. The crude toxins were extracted from toxic blue mussel with 80% ethyl alcohol and CH2Cl2, and the yield of toxin was 72.5%. GTX1, 4 and GTX2, 3 were purified from crude toxin by bio-gel P2 gel filtration column with Distilled water, 0.03 M AcOH and 0.05 M AcOH. The yield of toxin was 0.6%, 26.8% and 0% respectively. Bio-gel P2 gel filtration column with 0.03 M AcOH fraction toxin were purified from toxin by Bio-Rex 70 cation exchange column with 0.0-0.05 M AcOH (linear gradient). The toxin amount GTX1, 4 and GTX2, 3 were 840 ㎍ and 1,680 ㎍, respectively. The yield of GTX1, 4 and GTX2, 3 were 4.3% and 8.4%, respectively.
