Actinomycetes (223 strains) were isolated from marine sediments collected at various sites in the Jeollanam-do and their liquid culture media were analysed using HPLC for the production of new secondary metabolites. One of these actinomyces was isolated from solar saltern sediments collected at the Aphae-island. The strain, KMF-004 was cultured in an A1 liquid medium, and the production of secondary metabolites was monitored daily by HPLC-MS analysis. The HPLC chromatogram of 6 days culturing samples showed the presence of several notable secondary metabolites along with three previously reported bohemamine derivatives in the culture medium. These peaks were isolated and purified by flash column chromatography and prep-HPLC to yield five new compounds. Two novel compounds were identified with a new carbon skeleton. Their chemical structures were elucidated by intensive MS and 2D NMR study. Relative configuration was assigned by analyses of coupling constant in the 1H-NMR spectrum and NOE correlations in the 1D NOESY experiment. Antimicrobial activities of crude extract from KMF-004 were measured by agar diffusion assay. KMF-004 strain has shown weak antimicrobial activity to Micrococcus luteus and Enterococcus hirae. However 2FD12 and F10A2 strain has shown high antimicrobial activities to all test strains. These results have shown that are able to produce antimicrobial compounds. These results have shown that marine environment still contains a high chemical diversity and microbial diversity. In addition, the solar saltern-derived marine actinomycetes provide an attractive source for the discovery of novel secondary metabolites and to be potential candidates for antibiotics.
