Effect of Sub Minimal Inhibitory Concentration Antibiotics on the Morphology of Oral Bacteria

Antibiotics at sub-minimal inhibitory concentrations (sub-MICs) have been reported to affect bacterial morphology and biochemical characteristics. The purpose of this study was to examine the morphological changes induced in oral bacteria after treatment with sub-MICs of antibiotics. We treated cultures of Fusobacterium nucleatum, Porphyromonas gingivalis, Aggregatibacter actinomycetemcomitans, Actinomyces naeslundii, Actinomyces odontolyticus, Streptococcus gordonii, Streptococcus mutans and Lactobacillus acidophilus with sub-MICs of various antibiotics and analyzed the morphological changes induced in the bacteria by bright-field and scanning electron microscopy. The MICs for amoxicillin, doxycycline, metronidazole, penicillin, and tetracycline were determined by the broth dilution method. The lengths of A. actinomycetemcomitans and F. nucleatum increased after incubation with sub-MICs of metronidazole, penicillin, and amoxicillin, while that of P. gingivalis increased after incubation with sub-MICs of metronidazole and penicillin. The length of F. nucleatum decreased after incubation with sub-MICs of doxycycline and tetracycline. The lengths of A. naeslundii decreased after incubation with sub-MICs of penicillin and amoxicillin, while that of A. odontolyticus showed decreased length upon treatment with sub-MICs of doxycycline and tetracycline. L. acidophilus showed decreased length and thickness after incubation with sub-MICs of amoxicillin, doxycycline, penicillin, and tetracycline. Both S. gordonii and S. mutans showed increased size after incubation with sub-MICs of penicillin and amoxicillin. The bacterial chain length of S. mutans increased after incubation with sub-MICs of doxycycline and tetracycline. In conclusion, different antibiotics have distinct effects on the morphology of oral bacteria.
