This study was carried out to determine the effect of several in vitro culture conditions on the plantlet growth of two processing potato varieties, Dubaek and Atlantic. In order to get vigorous in vitro plantlets, some modifications in culture conditions such as membrane filter aeration system and concentration of sucrose and AgNO3 were applied and the growth and development of plantlets were examined. 1. Test groups with membrane filter aeration system showed higher CO2 and ethylene concentrations in vitro compared to the groups without membrane filter system. On the contrary, CO2 concentrations were measured lower in the groups with membrane filter aeration system. In vitro CO2 concentrations during the culture period in the test groups without membrane filter system increased sharply, while the ethylene concentrations decreased gradually. 2. Two weeks after transplanting to media, the survival rates of plantlets in the groups with membrane filter aeration system were over 70%, while those were only 11 to 19% in the groups without membrane filter system. 3. After culturing for 3 weeks, the medium remained in vitro reduced much more in the test groups with membrane filter aeration system than in the groups without membrane filter system. EC level was higher in the groups with membrane filter system and it was lowest when sucrose concentration was 3.0 to 4.5%. 4. Development of in vitro plantlets in the groups with membrane filter aeration system was much better than that of the groups without membrane filter system. Until sucrose concentration reached 3.0%, the growth of plantlets was quite good in both groups. But over 4.5% of sucrose concentration inhibited the growth of plantlets in both groups. The more sucrose level increased, the more the number of roots developed. But the length of root became shorter. When 3.0% sucrose was added, both of the dry weight and fresh weight were heavier in the groups with membrane filter aeration system than in the non-membrane filter system groups. 5. Chlorophyll contents in the groups with membrane filter aeration system were higher by 6.4 SPAD than those in the groups without membrane filter system. And Atlantic showed higher chlorophyll content by 5.6 SPAD as compared with Dubaek. 6. Although addition of AgNO3 inhibited the shoot length of in vitro plantlets, it showed good effect on the number of leaves and roots, the length of leaves and roots and chlorophyll content. When 20μM of AgNO3 for Dubaek and 10μM of AgNO3 for Atlantic were added, they showed the best results in the increase of chlorophyll content.
